A Process for Isolation and Purification of Curcuminoids from Curcuma longa
The present invention discloses a process for isolating a purified extract of curcuminoids from the rhizome of Curcuma longa. The process provides high yield of curcuminoids with purity more than 99%. The purified extracts of curcuminoids have anti-inflammatory, antioxidant, anti-tumor activity and may be used in pharmaceutical formulation thereof.

Area of Application: Curcuminoids are produced specifically by plants of the order Zingiberales and have long been used as food additives because of their aromatic, stimulant and colouring properties and as traditional Asian medicines because of their anti-tumor, antioxidant and hepatoprotective activities. A curcuminoid is a curcumin or a derivative of a curcumin with different chemical groups. Three main chemical compounds – Curcumin, Demethoxycurcumin and Bisdemethoxycurcumin are collectively known as Curcuminoids. A study evaluated the comparative antioxidant activity of curcuminoids in vitro using linoleic acid as the substrate in an ethanol/ water system as well as using rabbit erythrocyte membrane and rat liver. It was found that Tetrahydrocurcumin had the strongest antioxidant activity among all curcuminoids in each assay system. The results suggested that Tetrahydrocurcumin must play an important role in the antioxidant mechanism of Curcumin in vivo

Advantages: In the present invention, purity of the curcuminoid is estimated by HPLC. For HPLC analysis, both the standard extract (from Sigma) and the curcuminoids extracted from rhizomes of Curcuma longa are dissolved in mobile phase consisting of tetrahydrofuran and water in the ratio of 50:50. HPLC is carried out on C-18 column of 250 X 4.6mm with a flow rate of 1.0ml/min at a wavelength of 425nm.
