Unique Model for Screening Anti-Diabetic Compounds
The present technology provides a method for generating insulin non-responsive skeletal muscle cell culture model to screen anti-diabetic compounds against insulin resistance. The method comprises: A) Growing myocytes in serum containing medium. B) Differentiating the myocytes into myotubes in a mixture of serum free medium in the continuous presence of insulin. C) Determining the insulin-stimulated tyrosine phosphorylation of Insulin Receptor (IR) and/or Insulin receptor substrate-1 (IRS-1) myotubes differentiated in chronic presence of insulin to that of control (same cells differentiated in a mixture of serum free medium in the absence of insulin), wherein the reduced stimulation of tyrosine phosphorylation of IR and/or IRS-1 in myotubes differentiated in continuous presence of insulin as compared to control, indicated the development of insulin resistance, and D) screening the compounds to determine whether they are capable of enhancing insulin-simulated tyrosine phosphorylation of IR and/or IRS-1 myotubes differentiated in continuous presence of insulin to that of control (same cells are differentiated in the serum free medium in the absence of insulin).
