Protein coding RNAs to Correct Mitochondrial Dysfunction
The present technology provides a protein-coding RNA (pcRNA) useful for the treatment of mitochondrial dysfunction. The protein-coding correctional RNA used is a recombinant polynucleotide segment comprised of sequence operably linked to a signal tag. This signal-tagged RNA is administered as composition to the diseased subject with the help a protein carrier. The protein carrier helps in transporting the polynucleotide sequence across cellular and mitochondrial membranes. The signal-tagged correctional RNA upon reaching the mitochondria gets translated into functional protein. These functional proteins in turn correct the malfunctioning of mitochondria. The technology further suggests a kit with two active components, consisting of the carrier and the RNA, respectively, and a binding buffer, which are combined prior to use to form the ribonucleoprotein (RNP) complex. The RNP complex is administered through a suitable route. Further the use of the kit is for a diagnosis for screening mitochondria1 dysfunction, using the components provided in the kit. Statement of novelty The novel features of the present technology are: 1)It delivers large RNAs of many kilobases length, while the existing technologies describes the delivery of short RNAs (-40 bases). 2)The pcRNAs of this technology have protein-coding sequences which, upon expression within mitochondria, are translated to functional proteins. 3)pcRNAs stimulate respiration in cells bearing mitochondria1 deletions. 4)A combination of pcRNAs with genome-wide coverage of protein-coding sequences is used to correct genetic defects. 5)It employs functional carrier complexes R6 and R8 which are expressed in bacteria and assembled in vitro.
