A Novel Polysaccharide Anti-Cancer Agent
The invention in discussion, relates to a novel bioactive polysaccharide isolated from edible mushroom, Agaricus bisporus. The invention also relates to a pharmaceutical composition comprising the bioactive polysaccharide and provides steps of the process for isolation of the bioactive polysaccharide. There have been extensive studies demonstrating the anti-cancer activity of the extracted, purified glucan polysaccharides (Wasser & Weis 1999). From studies on various cancer cell lines, there is evidence for direct cytotoxic effects on the cancer cells for some of the polysaccharides. Highly encouraging results have been forthcoming together with the observations that the incorporation of bioactive polysaccharides into treatment regimes significantly reduced the side-effects so often encountered bypatients. These compounds can increase the quality of life of cancer patients and may offer increased survival rates for some types of cancer. The invention deals with a new molecule, which is a potential candidate against cancer. This molecule is a polysaccharide which is partially fucosylated galactoglucans. The compound isolated by the inventors is unique in that it consists of three sugars glucose, galactose and fucose linked in a specific manner, in a specific ratio and as a two-dimensional structure. The bioactivity of the technology is better than lentinan and krestin, the commercially available polysaccharides tested at the same concentration. In order to avoid problems faced in obtaining polysaccharides like even after tedious procedures the polysaccharide obtained is very less, is not pure and is almost always contaminated, the inventors have developed a process whereby polysaccharides may be obtained in less than 48 hours. The present process employs very few steps, and uses harmless materials like NaCl for dialysis. The dialysate can be directly used to obtain the final compound in purity of >98%. In fact, from l kg of Agaricus material, 160 mg of compound can be obtained. The invention provides pharmaceutical compositions comprising the novel polysaccharide compound of the invention as active ingredient, together with pharmaceutically acceptable additives. The composition may be prepared in any physical form such as tablet, syrup or capsules.
